Summary

A novel approach for biomaterial-mediated enhancement of vaccination was developed based on lymph node injection of adjuvant-loaded microparticles. A single injection of these materials elicited antigen-specific CD8+ T-cell responses that were dramatically enhanced compared with injection of soluble formulations, and the magnitude of these responses were on the order of those typically associated with viral vectors. Mechanistic studies reveal CD8+ T-cell enhancement is driven by increased adjuvant retention within the lymph node (LN) and that this retention promotes more enduring DC activation.
