Investigation of Piracetam Cocrystals Formation During Different Grinding Procedures by Raman Spectroscopy

Summary

The aim of this study was to examine the applicability of Raman spectroscopy in combination with chemometrics for the investigation of piracetam cocrystal formation during different grinding procedures. As a model API, polymorphic forms I and III of piracetam were used. Cocrystal former was L-tartaric acid. It could be shown that physical mixing did not succeed in forming cocrystals. On the other hand, cocrystals were formed within 1 min by solvent drop grinding. Raman spectroscopy revealed time-dependent cocrystal formation of piracetam/L-tartaric acid during dry-grinding at the applied milling times.
